What other options are there for travelling today, even with a layover?
No me sirve — it doesn't work for me — rejects the offer without rejecting the person, and it is the phrase that reopens the search. Say the constraint once, plainly: tengo que estar allí esta noche. Everything the agent finds after that is found against your deadline, not theirs.
The airline's first offer is built for the airline: tomorrow morning, next available, please step aside. Accepting it takes one word. Not accepting it — politely, in Spanish, with a queue shifting behind you — is a negotiation under time pressure, which is why it sits at the top of the course.

Two things worth knowing
The first offer is rarely the only one in the system.
No me sirve is a key, not a complaint
It sounds blunt translated — "that doesn't serve me" — but in Spanish it is the neutral, standard way to reject an option while keeping the conversation going. It carries no accusation, so the agent does not have to defend the cancelled flight; they just go back to the screen. Follow it with the constraint and you have redefined what counts as a solution.
Say the new plan back before you walk away
«Entonces: salida hoy con escala en Lisboa, llegada sobre medianoche» takes ten seconds and closes the two gaps a rebooking leaves open — the ones between what you think was booked, what the agent thinks you accepted, and what the system recorded. If those three ever disagree, they disagree at midnight, in an airport, in Spanish. The repeat-back is where you catch it instead.
